MCF-7 cell transplantation models

Breast adenocarcinoma (ER+)

Classic hormone-responsive breast line (Soule 1973). s.c. (Kang 6×10⁶) and #4 fat-pad (Dall 1×10⁶) usually need exogenous estrogen; OVX without a pellet can fail completely. Doses are not interchangeable across routes, and MDA-MB-231 numbers do not copy over. RUO.

MCF-7 subcutaneous xenograft

ER+ MCF-7 flank/forearm s.c. growth usually needs exogenous estrogen and Matrigel for endocrine pharmacology. Kang: 6×10⁶ in ice-cold Matrigel. Not fat-pad orthotopic and not a mets model. Do not copy MDA-MB-231’s 2×10⁶ / 100 μL.

MCF-7 orthotopic mammary fat-pad tumor

#4 fat-pad ER+ MCF-7 typically needs exogenous estrogen. This SOP uses Dall: 1×10⁶ in PBS + GFR Matrigel 1:1. Shafie: 2×10⁶ takes in intact nude fat pads; OVX fails even at 20×10⁶. Watch the fat-pad JoVE—not a substitute for surgical training. Do not copy MDA-MB-231’s 2×10⁶ / 100 μL / 25% matrix.
